最初の１点目については、

25/tcp : メールサーバー間の基本的な通信プロトコル
465/tcp or 587/tcp : メールクライアントがメールサーバーへメール送信する際に利用するプロトコル
pop3やIMAPはメールクライアント側がメールサーバーに蓄積されたメールを受信するためのプロトコル

と理解したらよいと思います。つまりは外部からメールを配送してもらったり、こちらからメールを外部へ配送するのには
25/tcpをつかうという約束事があるということです。従って外部とのメール送受信する際には25/tcpをあけておく必要があります。

ただ、SMTPの仕組みを理解しないと、トラブル起こった時にやばそうには思います。
２番目はMXレコードが正しいかどうか、外部からtelnetなどを使って直接SMTPコマンドをたたいてメール送信を試してみてOKかどうか

ぐらいのチェックは基本かなと思います。
